Research on NMN includes cell studies, animal experiments, and a growing number of human trials. Many early findings come from mice, where changes in NAD+ levels and metabolic markers have been reported. Human data are more limited, and questions remain about effective routes of administration, tissue distribution, and long-term effects. Some trials measure NAD+ in blood or tissue, while others assess physical function or metabolic outcomes. Regulatory status differs between countries, and NMN is not universally approved as a dietary supplement or therapeutic agent.
Nicotinamide mononucleotide, commonly abbreviated NMN, is a naturally occurring nucleotide. Its structure combines a nicotinamide ring with a ribose sugar and a phosphate group. The compound appears in cells across many organisms as an intermediate in the production of nicotinamide adenine dinucleotide, or NAD+. Because NMN sits close to NAD+ in metabolism, it has drawn interest in biochemistry and aging research. The molecule is not a dietary essential nutrient in the classical sense, and its presence in food is generally low and variable.
NAD+ serves as a coenzyme in redox reactions and as a substrate for enzymes involved in DNA repair and cellular signaling. In the salvage pathway, nicotinamide is converted to NMN by the enzyme NAMPT. NMN is then converted to NAD+ by NMNAT enzymes. A separate route links nicotinamide riboside to NMN through phosphorylation. These pathways maintain NAD+ levels, which can decline with age or metabolic stress in some tissues. The relative contribution of circulating NMN to tissue NAD+ remains an active area of study.
Commercial NMN is produced through enzymatic or chemical routes, and the resulting material can vary in purity, counterion, and residual solvent content. Buyers typically rely on certificates of analysis, but independent verification through third-party laboratories provides stronger assurance. Regulatory treatment differs by country; in the United States, NMN has been subject to shifting guidance about its status as a dietary supplement, while other markets permit sales under local rules. No universal pharmacopeial monograph exists for NMN, so specifications often come from suppliers, research protocols, or regional requirements.
Solid NMN is generally handled as a moisture-sensitive compound. Dry material stored desiccated at low temperature, protected from light, tends to remain stable for extended periods. Aqueous solutions are less stable and can undergo hydrolysis, especially at elevated temperature or alkaline pH. The anomeric form also matters: beta-NMN is the naturally occurring form, while alpha-NMN can appear as a synthetic impurity. Purity and storage conditions therefore influence both analytical results and experimental reproducibility.

Inside cells, the enzyme nicotinamide phosphoribosyltransferase, or NAMPT, converts nicotinamide and a ribose-phosphate donor into NMN. A second enzyme, NMN adenylyltransferase, then converts NMN into NAD+. NAD+ participates in redox reactions and serves as a substrate for signaling enzymes such as sirtuins, PARPs, and CD38. Because NAD+ levels tend to decline with age in many organisms, NMN has drawn interest as a possible way to influence that decline. Whether oral NMN reliably raises NAD+ in human tissues, and whether any such change modifies disease risk, remain open research questions.
NMN is present in small amounts in some foods, including certain vegetables, fruits, and animal products, but food content varies widely and is not well standardized. In laboratory research, NMN is used as a tool compound to study NAD+ metabolism, mitochondrial function, and cellular stress responses. Animal studies have reported changes in NAD+ levels and various physiological measures after NMN administration, but species differences and study designs limit direct extrapolation to humans. Human trials have largely focused on safety, tolerability, and pharmacokinetics, with fewer studies examining clinical endpoints.
Nicotinamide mononucleotide, commonly abbreviated NMN, is a naturally occurring nucleotide. Its structure consists of a nicotinamide ring attached to a ribose sugar that carries a phosphate group. The molecular formula is C11H15N2O8P, and the molar mass is about 334.22 grams per mole. In cells, NMN is an intermediate in the salvage pathway that recycles nicotinamide to maintain NAD+ levels. It is not the same compound as NAD+, although it is a direct precursor in one enzymatic step.
Quantifying NMN requires methods that separate it from structurally similar compounds such as nicotinamide, nicotinamide riboside, and NAD+. Common approaches include high-performance liquid chromatography coupled with ultraviolet detection, liquid chromatography with tandem mass spectrometry, capillary electrophoresis, and nuclear magnetic resonance for identity confirmation. Because NMN is polar and often present at low concentrations in biological samples, sample preparation can involve protein precipitation, solid-phase extraction, or derivatization. Isotope-labeled internal standards help correct for matrix effects and recovery losses. Reported concentrations depend heavily on the matrix, extraction protocol, and analytical platform.
Stability of NMN depends on physical form, temperature, moisture, light, and pH. The solid compound is generally more stable than aqueous solutions, which can degrade over time, especially when warm or exposed to extreme pH. Recommended laboratory storage is typically desiccated at −20 °C or below, protected from light, with containers sealed to limit moisture uptake. In solution, degradation products may include nicotinamide and related ribosides, and the rate varies with buffer composition and concentration. Analytical laboratories often prepare fresh solutions and validate stability for each method.
=== Prochirality and three-point attachment === The concept of prochirality is necessary for understanding some aspects of enzyme stereospecificity. Ogston pointed out that when a symmetrical molecule is placed in an asymmetric environment, such as the surface of an enzyme, supposedly identically placed groups become distinguishable. In this way he showed that earlier exclusion of non-chiral citrate as a possible intermediate in the tricarboxylate cycle was mistaken. A coffee mug with one handle is an everyday example of a prochiral object. If it is placed in an achiral corrosive liquid, such as a concentrated acid, then the left and right-hand sides will be corroded equally because there is nothing to distinguish them. However, if the mug is held in a person's right hand it is easy to drink out of the left-hand side but difficult to drink out of the right-hand side. In other words, an achiral environment such as an acid cannot distinguish between the two sides of a prochiral, but a chiral object like a person can. Thus a chiral enzyme such as aconitase can act differently on two apparently equivalent groups on a prochiral molecule, so citrate can be an intermediate in the tricarboxylate cycle.
== Research == Tetracosactide is a corticosteroid that causes the brain to produce additional spinal fluid to replace the volume of the lost CSF and alleviate symptoms, and has been given intravenously to treat CSF leaks. In three small studies of 1-2 patients with recurrent CSF leaks where repeated blood patches failed to form clots and relieve symptoms, the patients received temporary but complete resolution of symptoms with an epidural saline infusion. The saline infusion temporarily restores the volume necessary for a patient to avoid SIH until the leak can be repaired properly. Intrathecal saline infusion is used in urgent cases such as intractable pain or decreased consciousness. The gene TGFBR2 has been implicated in several connective tissue disorders including Marfan syndrome, arterial tortuosity, and thoracic aortic aneurysm. A study of people with a sCSF leak demonstrated no mutations in this gene. Minor features of Marfan syndrome have been found in 20% of CSF leak patients. Abnormal findings of fibrillin-1 have been documented in these CSF leak patients, but only one patient demonstrated a fibrillin-1 defect consistent with Marfan syndrome.
=== Pregnancy and lactation === Use of tramadol during pregnancy is generally avoided, as it may cause some reversible withdrawal effects in the newborn. A small prospective study in France found, while an increased risk of miscarriages existed, no major malformations were reported in the newborn. Its use during lactation is also generally advised against, but a small trial found that infants breastfed by mothers taking tramadol were exposed to about 2.88% of the dose the mothers were taking. No evidence of this dose harming the newborn was seen.

Clinical metagenomic next-generation sequencing (mNGS) is the comprehensive analysis of microbial and host genetic material (DNA or RNA) in clinical samples from patients by next-generation sequencing. It uses the techniques of metagenomics to identify and characterize the genome of bacteria, fungi, parasites, and viruses without the need for a prior knowledge of a specific pathogen directly from clinical specimens. The capacity to detect all the potential pathogens in a sample makes metagenomic next generation sequencing a potent tool in the diagnosis of infectious disease especially when other more directed assays, such as PCR, fail. Its limitations include clinical utility, laboratory validity, sense and sensitivity, cost and regulatory considerations. Outside of clinical medicine, similar work is done to identify genetic material in environmental samples, such as ponds or soil.

=== Kyrgyzstan === Kyrgyzstan has a similar HIV/AIDS problem as Ukraine and to try and combat this, started a trial methadone program within prisons. The first program started in a solo prison is 2002 and expanded to multiple in 2008. It is estimated that half of incarcerated individuals in Kyrgyzstan inject drugs and the use of unsterilized needles and transfer of HIV is higher in the prison setting. While the offering of methadone in prisons would help curb the spread of HIV/AIDS, the internal prison governance of inmates and their reliance on the drug trade is a hard barrier. Heroin being used as currency as well as methadone users being assigned to separate living quarters than non-users creates a social separation between users and non-users. These outcasting factors can make methadone treatment not worth it for some inmates. The number of eligible prisoners using methadone through a methadone program is estimated at only 7%.
== History == The efficacy of ceftolozane/tazobactam to treat complicated intra-abdominal infections (cIAI) in combination with metronidazole was established in a clinical trial with a total of 979 adults. Participants were randomly assigned to receive ceftolozane/tazobactam plus metronidazole or meropenem. Results showed ceftolozane/tazobactam plus metronidazole was effective for the treatment of cIAI. The efficacy of ceftolozane/tazobactam to treat complicated urinary tract infections (cUTI) was established in a clinical trial where 1,068 adults were randomly assigned to receive ceftolozane/tazobactam or levofloxacin. Ceftolozane/tazobactam demonstrated it was effective in treating cUTI. Ceftolozane/tazobactam was shown to be at least as effective as other antibiotics in curing infections in three main studies. One study involved 1,083 participants who mostly had kidney infection or in some cases a complicated urinary-tract infection. Ceftolozane/tazobactam successfully treated the infection in about 85% of the cases where it was given (288 of 340), compared with 75% (266 of 353) of those given another antibiotic called levofloxacin. The second study involved 993 participants with complicated intra-abdominal infections. Ceftolozane/tazobactam was compared with another antibiotic, meropenem. Both medicines cured 94% of participants (353 out of 375 given ceftolozane/tazobactam and 375 out of 399 given meropenem). The third study involved 726 participants who were using a ventilator and who had either hospital-acquired pneumonia or ventilator-associated pneumonia.
=== Toxicity of pentavalent arsenicals === Due to its similar structure and properties, pentavalent arsenic metabolites are capable of replacing the phosphate group of many metabolic pathways. The replacement of phosphate by arsenate is initiated when arsenate reacts with glucose and gluconate in vitro. This reaction generates glucose-6-arsenate and 6-arsenogluconate, which act as analogs for glucose-6-phosphate and 6-phosphogluconate. At the substrate level, during glycolysis, glucose-6-arsenate binds as a substrate to glucose-6-phosphate dehydrogenase, and also inhibits hexokinase through negative feedback. Unlike the importance of phosphate in glycolysis, the presence of arsenate restricts the generation of ATP by forming an unstable anhydride product, through the reaction with D-glyceraldehyde-3-phosphate. The anhydride 1-arsenato-3-phospho-D-glycerate generated readily hydrolyzes due to the longer bond length of As-O compared to P-O. At the mitochondrial level, arsenate uncouples the synthesis of ATP by binding to ADP in the presence of succinate, thus forming an unstable compound that ultimately results in a decrease of ATP net gain. Arsenite (III) metabolites, on the other hand, have limited effect on ATP production in red blood cells.
Most of the insurgents apparently concealed their arms and vanished into the local population. This proved less successful in Zambia, where the civilians in Sesheke District were irritated by the constant presence of South African patrols and reconnaissance aircraft; they demanded their government remove the remaining PLAN fighters. President Kaunda subsequently bowed to pressure and ordered PLAN to close its rear base facilities in Zambia, resulting in the collapse of its Caprivi insurgency. On 16 March, Angola lodged a formal complaint with the UN Security Council concerning the violation of its borders and airspace as a result of Operation Rekstok. United Nations Security Council Resolution 447 was passed in response. The resolution "condemned strongly the racist regime of South Africa for its premeditated, persistent, and sustained armed invasions of the People's Republic of Angola, which constitute a flagrant violation of the sovereignty and territorial integrity of the country as well as a serious threat to international peace and security". A UN commission of inquiry logged 415 border violations by the SADF in 1979, an increase of 419% since the previous year. It also made note of 89 other incidents, which were mostly airspace violations or artillery bombardments that struck targets on Angolan soil.

== Early life and education == Larsen earned a BS in chemistry from Santa Clara University in 1978, and a PhD in chemistry under the direction of Douglas P. Ridge from University of Delaware in 1983. She completed her post-doctoral research with Catherine Fenselau at Johns Hopkins University.
A review of lichen taxonomic literature from 2018 to 2020 found that of over 700 new species published, only 39% included any DNA sequences. The most commonly used gene was the ITS (present in roughly 82% of those that had molecular data), while only about 10% of new species were supported by three or more genes. These figures show that while multilocus sequencing underpins higher-level systematics, species-level descriptions (alpha taxonomy) often remain constrained by practical limits on sequencing or by the sufficiency of morphological evidence. Sanger-era phylogenetics laid the groundwork for later genomic studies. By the late 2000s lichenologists had a working framework for most major lineages and clearer criteria for natural versus artificial groups. The framework relied on what now seem small datasets—only a few kilobases per species—yet these sequences resolved many relationships. Although some 2010-era authors questioned the value of small multigene matrices, Lücking (2020) contends that sound sampling and analysis can outweigh sheer data volume. By the early 2020s, many new species—even some higher taxa—are still described from a few gene regions plus morphology, a practise that remains practical where large-scale sequencing is not yet feasible. The Sanger era showed that modest molecular datasets could overturn classifications—splitting some genera, merging others—and it supplied a scaffold for later genome-scale studies.
TNF is a cytokine produced mainly by activated macrophages, and is the major extrinsic mediator of apoptosis. Most cells in the human body have two receptors for TNF: TNFR1 and TNFR2. The binding of TNF to TNFR1 has been shown to initiate the pathway that leads to caspase activation via the intermediate membrane proteins TNF receptor-associated death domain (TRADD) and Fas-associated death domain protein (FADD). cIAP1/2 can inhibit TNF signaling by binding to TRAF2. FLIP inhibits the activation of caspase-8. Binding of this receptor can also indirectly lead to the activation of transcription factors involved in cell survival and inflammatory responses. However, signalling through TNFR1 might also induce apoptosis in a caspase-independent manner. The link between TNF and apoptosis shows why an abnormal production of TNF plays a fundamental role in several human diseases, especially in autoimmune diseases. The TNF receptor superfamily also includes death receptors (DRs), such as DR4 and DR5. These receptors bind to the protein TRAIL and mediate apoptosis. Apoptosis is known to be one of the primary mechanisms of targeted cancer therapy. Luminescent iridium complex-peptide hybrids (IPHs) have recently been designed, which mimic TRAIL and bind to death receptors on cancer cells, thereby inducing their apoptosis.
